Dansyl-Glu-Gly-Arg-chloromethylketone is a fluorescent substrate for epidermal growth factor (EGF). It has been shown to inhibit the proteolytic activity of EGF. This inhibition is due to the binding of Dansyl-Glu-Gly-Arg-chloromethylketone to calcium and its inhibitory properties on EGF. The flow system used in this experiment was activated with a monoclonal antibody, which binds to the cytosolic calcium and activates the enzyme. The kinetic data obtained from this experiment showed that the rate of EGF hydrolysis by Dansyl-Glu-Gly-Arg-chloromethylketone was proportional to the concentration of calcium in the cytosol.
