Diethyl isocyanatidophosphate (DI) is a novel, non-competitive inhibitor of mammalian cell surface transporters that has been shown to inhibit the uptake of glucose and other nutrients. DI binds reversibly to the transporter, blocking it from transporting glucose across the plasma membrane. This has been shown to induce weight loss in obese rats with a body mass index greater than 30%. DI’s mechanism of action is similar to that of macrocyclic inhibitors, which act by binding to the ATP-binding site on mammalian cell surface transporters. The binding prevents ATP from being released and thus blocks substrate uptake. DI has also been shown to block bacterial pathogens such as methicillin resistant Staphylococcus aureus and Mycobacterium tuberculosis, which are both responsible for serious diseases including pneumonia and tuberculosis.
