3,5-Diamino-1,2,4-triazole is a nitrogen heterocyclic compound that has shown to be an inhibitor of the Toll-like receptor (TLR) signaling pathway. The TLR pathway is a major regulator of the innate immune system and plays a role in the development of many infectious diseases. 3,5-Diamino-1,2,4-triazole has been shown to inhibit bacterial growth in model systems and leukemic mice. It is also shown to have high binding affinity for TLRs. 3,5-Diamino-1,2,4-triazole binds to the Toll/interleukin 1 receptor domain (TIR) by hydrogen bonding interactions with amino acid residues and competitively inhibits the binding of ligands such as lipopolysaccharide (LPS) or peptidoglycan.
