The synthesis of (2-(dimethyl(4-((4-methylpiperazin-1-yl)methyl)phenyl)silyl)phenyl)methanol was achieved in a stereoselective manner. The chiral alcohol was prepared in two steps from commercially available starting materials. The first step is the oxidation of the vinyl group to the corresponding radical, followed by the cyclization of this radical to afford a diastereomeric mixture of 1,2-dideoxy alcohols.
The second step involves the reaction of these alcohols with bromomethylbenzene and potassium fluoride in methanol to afford the desired product in high yield and excellent optical purity.
