2,3-Diamino-5-bromopyridine (2,3-DBP) is a unsymmetrical diamine that can be used as a cancer therapeutic. 2,3-DBP inhibits the activity of multidrug efflux pumps in cancer cells by binding to the transporter protein and inhibiting its function. This drug also inhibits glycogen synthase kinase-3 (GSK-3), which is an enzyme with tumor suppressor properties. In addition, 2,3-DBP is cytotoxic to cancer cells in vitro and has been shown to inhibit tumor growth in vivo. 2,3-DBP was shown to have an IC50 of ~8 μM for the efflux pump MDR1 and ~4 μM for Pgp. It also showed cytotoxicity against a panel of human cancer cell lines at concentrations ranging from 10 nM to 1 μM.
