2-Ethyl-6-hydroxypyrimidin-4(3H)-one is an intermediate in the synthesis of the drug propionamide. It can be prepared by reacting ethyl pyrimidine-2,4(1H,3H)-dione with diethyl chloroacetate in the presence of a catalyst such as sodium methoxide. The reaction time can vary from one to three hours and the optimal reaction temperature is between 50°C and 60°C. 2-Ethyl-6-hydroxypyrimidin-4(3H)-one can also be prepared using propionitrile and alkali solution. This process takes approximately four hours to complete, but yields a greater quantity of product than when using diethyl chloroacetate.
