Fast Blue BB salt is a fluorescent dye that is used in histology and cytology to identify cytoplasmic structures. It can be used for the detection of basic proteins, such as actin, in tissue culture. Fast Blue BB salt reacts with acidic groups in the protein to form a blue-green chromophore. This reaction mechanism is similar to that of Methylene Blue, which also reacts with acidic groups on proteins to form a blue-green chromophore. The biological sample may be activated by heating or by treatment with enzymes or detergents before staining with Fast Blue BB salt. The polymerase chain reaction (PCR) technique has been used to amplify DNA sequences using Fast Blue BB salt as a fluorogenic probe. The fluorescence of this dye is proportional to its concentration and it is therefore suitable for quantification when conducting PCR reactions.
