2-(Fluoromethyl)-Ornithine (2-FMEO) is a potent antitumor agent that is an irreversible inhibitor of polyamine biosynthesis. It has been shown to inhibit the growth of cancer cells and other tumor cells in experimental models. 2-FMEO inhibits the enzyme ornithine decarboxylase, which converts ornithine to putrescine, by binding reversibly to the nitrogen atom in the active site. This inhibition leads to a depletion of polyamines, which are necessary for cell growth. 2-FMEO also inhibits spermidine synthase and spermidine/spermine N1-acetyltransferase, which are enzymes involved in the synthesis of spermidine and spermine from putrescine.
