12alpha-Hydroxyrotenone is a naturally occurring compound that has been shown to have significant cytotoxicity against cultured cells and human serum. 12alpha-Hydroxyrotenone has been found to be an inhibitor of the enzyme dinucleotide phosphate (NADPH) oxidase, which is involved in the production of reactive oxygen species. This inhibition may be due to its ability to act as an antioxidant by donating hydrogen atoms to free radicals or through its hydroxyl group. 12alpha-Hydroxyrotenone also binds to cell membranes, which may account for some of its cytotoxicity. The mechanism of cellular uptake and subsequent intracellular degradation is not known.
