5-Hydroxymethyldeoxyuridylate is a nucleotide analog that is used to treat cancer. It inhibits the synthesis of DNA by binding to the enzyme DNMT1 and causing it to lose its ability to produce DNA. 5-Hydroxymethyldeoxyuridylate also inhibits the synthesis of RNA, which leads to cell death. This drug has been shown to be effective in treating both bacterial and mammalian cells. 5-Hydroxymethyldeoxyuridylate is active against a range of bacteria, such as subtilis, and can cause genetic ablation in mammalian cells. The latter property may be due to its ability to bind with adenine residues on the DNA strand and block them from being incorporated into new strands. 5-Hydroxymethyldeoxyuridylate binds reversibly with DNA at neutral pH, which allows it to be removed from the cell by washing or other means.
