4-Iodo-5-methyl-1H-imidazole is a primer that is used in diagnostic PCR. Primers are short DNA or RNA molecules that are complementary to the sequence of a gene or part of a gene. 4-Iodo-5-methyl-1H-imidazole is used as a primer for polymerase chain reaction amplification of acid sequences in DNA and RNA. It has a sensitivity of 10 copies per reaction, which makes it useful for detecting cancer cells. The primers bind to the DNA template by hydrogen bonding, which prevents the extension of the growing strand by the polymerase enzyme. This prevents an exponential increase in nucleotide synthesis and results in an arithmetic progression of nucleotides incorporated into the new strands.
