N-alpha-Z-D-lysine is a colorimetric method for detecting the presence of lysine residues in proteins. It is based on the reaction of lysine residues with pipecolic acid, which yields a red product. The shift of the nmr spectrum of N-alpha-Z-D-lysine is used to determine the number of lysine residues in a protein. This method has been used to detect lysine residues in erythrocyte membranes and photoreceptors, as well as to study the structure of proteins. N-alpha-Z-D-lysine can be desalted by ion exchange chromatography and has been shown to produce high yields from bovine tissues.
