5,10-Methylenetetrahydrofolate is a dinucleotide phosphate that is the precursor to tetrahydrofolate (THF), which is required for the synthesis of DNA. 5,10-Methylenetetrahydrofolate is synthesized from dihydrofolate (DHF) and formaldehyde through two reactions catalyzed by enzymes formyltetrahydrofolate synthase and dihydrofolate reductase. The enzyme formyltetrahydrofolate synthase uses 5,10-methylenetetrahydropteroyltriglutamate as a substrate and produces 5,10-methylenetetrahydrofolic acid.
