Methanesulfonylacetone is a nitro-containing alkylating agent that reacts with amides to form an active methylene group. It is prepared by the reaction of methanesulfonic acid with acetone in the presence of phosphorus oxychloride. Methanesulfonylacetone has been shown to inhibit hypertension in rats and to have antihypertensive activity. Methanesulfonylacetone also showed reversible covalent binding to DNA, which may be due to its ability to react with nucleophilic groups on the aromatic rings of DNA bases. Methanesulfonylacetone also showed anticancer properties, which may be related to its ability to cross the blood-brain barrier and inhibit protein synthesis in cancer cells.
