3-Nitrosopyridine-2,6-diamine is a fluorometric assay for the detection of nitrite in urine. The reaction mechanism is based on the reaction of 3-nitrosopyridine-2,6-diamine with nitrite to form a fluorescent product that can be measured using a fluorometer. This assay is sensitive and selective, and has been used to measure nitrite levels in biological fluids such as urine and saliva. 3-Nitrosopyridine-2,6-diamine was first described by German chemist Fritz Haber in 1907. In this experiment, Haber reacted the chemical with potassium hydroxide (KOH) to produce an acid solution that contains nitrite ions. When mixed with water containing an indicator dye such as phenolphthalein or methyl red, it produces a pink color that can be quantified using the fluorescence technique.
