NBD hexanoic acid is a fluorescent probe that is used as a substrate for measuring the activity of phospholipases. It is activated by the enzyme phospholipase A2, which cleaves off the fatty acid at the sn-2 position. This reaction is followed by hydrolysis of NBD hexanoic acid to release a chromophoric product that has a maximum excitation wavelength of 350 nm and an emission wavelength of 450 nm. The rate of hydrolysis can be determined by measuring the fluorescence intensity at different time intervals using a fluorimeter. The rate constant for this reaction can be obtained from the slope of the linear regression line.
