Netobimin is a bicyclic heterocycle that has been shown to have potent anthelmintic activity. It can be used in the treatment of intestinal nematodirus infections and other helminthiasis. Netobimin is administered orally, with an effective dose of 400 mg, and has a complex mechanism of action. It inhibits the glutamate dehydrogenase enzyme in the nematode and prevents it from synthesizing glutamate, which causes paralysis. Netobimin also binds to benzimidazole compounds that are present in the worm’s body, preventing them from binding to GABA receptors on the surface of nerve cells. This binding prevents the formation of a chloride channel between nerve cells, resulting in muscle paralysis. Netobimin also inhibits fatty acid synthesis by blocking acetyl-CoA carboxylase (ACC) and carnitine palmitoyltransferase I (CPT-I).
