Pristinamycin IA is a macrolide antibiotic that has been found to be effective against gram-positive bacteria, including Streptococcus pyogenes, S. pneumoniae, and Staphylococcus aureus. It also has synergistic effects when used with antibiotics such as tetracycline or erythromycin. Pristinamycin IA inhibits the synthesis of bacterial protein by binding to the 50S ribosomal subunit, as well as inhibiting DNA gyrase and topoisomerase IV. The discovery process for this drug was based on the observation that a mutant strain of Streptomyces coelicolor (strain A) was resistant to streptogramins. This led to the discovery that these mutants have an increased resistance to pristinamycin IA due to mutations in their 16S ribosomal RNA.
