Descripción

Picrotin is a synthetic analogue of the natural product picrotoxinin. It is a potent inhibitor of the influenza virus polymerase, which prevents viral replication. Picrotin also has pharmacological activity in autoimmune diseases and as an analgesic. The active analogues of picrotoxinin bind to the α1 subunit of the enzyme, which prevents the formation of a functional polymerase chain reaction. This binding prevents transcription and replication by inhibiting DNA synthesis and protein synthesis. Picrotin binds to carbonyl oxygens on proteins and forms disulfide bonds with other thiols such as glutathione, cysteine, or cystine to form a complex that binds to the agonist binding site on target enzymes.
