1h-Pyrrolo[2,3-b]pyridin-5-ol is a synthetic chemical that is used as an intermediate in the synthesis of the drug venetoclax. It has been shown to be an inhibitor of Bcl-2 and is a fluorine-containing heterocycle. The chemical is synthesized by reacting 2,4,6-trifluoroacetic acid with 1H-pyrrole in the presence of Grignard reagent and trimethylchlorosilane. The reaction intermediates are then purified and converted into 1h-pyrrolo[2,3-b]pyridin-5-ol using trifluoroacetic acid and hydrochloric acid. Impurities can be removed by recrystallization or distillation. 1h-Pyrrolo[2,3-b]pyridin-5-ol is industrially manufactured by reacting
