The in vivo activity of 8-fluoro-1-naphthoic acid has been shown to be CB2 receptor mediated. It was found to have a high affinity for the CB2 receptor, with an EC50 value of 0.5 nM. The electron affinities and substituents were also found to be important for the binding of 8-fluoro-1-naphthoic acid to the CB2 receptor, with bromo substituents being most effective.
8-Fluoro-1-naphthoic acid is an analog of 2-(4'-chlorobiphenyl)-N-(piperidin-1-yl)ethanamine (JWH133), which binds with high affinity to the cannabinoid receptor CB2 and is a potent agonist at this site, but not at the cannabinoid receptor CB1. 8FNA acts as an agonist at both receptors, but it has higher affinity for the
