Tolebrutinib is being developed by Novartis as a monoclonal antibody, which binds to the target protein Bruton's tyrosine kinase and inhibits its function. The binding of tolebrutinib with the protein prevents it from phosphorylating other proteins, thereby inhibiting their function. The binding of tolebrutinib with the protein also induces reversible covalent modifications at a number of sites on the protein, which may lead to the development of resistance over time.
