Thieno[3,2-b]pyridin-7-ol is a methoxylated analog of the nucleoside thymidine. It is programmed to bind to DNA in a sequence specific manner and can be used for the treatment of lung cancer. Thieno[3,2-b]pyridin-7-ol binds to the pBR322 plasmid DNA by an intercalative mechanism. This binding prevents replication and transcription, which leads to cell death. Thieno[3,2-b]pyridin-7-ol has been shown to have a cytotoxic effect on human lung cancer cells in culture. Studies have shown that this drug inhibits tumor growth in mice with breast cancer and rat primary cells with prostate cancer.
