A thrombin receptor agonist is a substance that binds to the thrombin receptor and activates it. This is a paradigm shift in the understanding of how the thrombin receptor works, as previously it was thought that the receptor only responded to extracellular stimuli. The most studied example of a thrombin receptor agonist is clopidogrel, which has been shown to activate the receptor by binding to it and stimulating its signaling pathways. Clopidogrel also inhibits platelet aggregation by preventing the formation of prothrombinase complexes with platelets. Clopidogrel is used as an anti-platelet agent and works by blocking adenosine diphosphate (ADP) receptors on platelets. It also prevents activation of protein C, which is an anticoagulant enzyme produced in response to tissue injury or inflammation.
