AICAR phosphate is a metabolite of AICAR that is an AMP-activated protein kinase (AMPK) activator. It was originally developed as an anti-obesity drug, but its use has been limited due to the high incidence of side effects. AICAR phosphate has been shown to be effective against inflammatory bowel disease in animal models by reducing the tissue inflammation and restoring tissue integrity. It also inhibits histidine decarboxylase and reduces levels of nucleotides, which are precursors to inflammatory cytokines. The exact mechanism of action remains unknown, but it appears that this drug may block the activation of NOD-like receptor pyrin domain 3 (NLRP3) inflammasomes in epithelial cells.
