N-Benzylglycine tert-butyl ester is an azomethine-containing molecule that has shown to be a potent and selective antagonist of the gonadotropin-releasing hormone receptor. This drug has been shown to be a competitive inhibitor of the 1,3-dipolar cycloaddition reaction between N-(benzyloxycarbonyl)glycine and tertiary butyl diazoacetate. The pharmacophore for this drug is derived from the carboxylic acid moiety at C2, which is necessary for binding to the receptor, and the benzyl group at C4 that is essential for binding to the enzyme. The stereoselectivity of this compound was determined by X-ray crystallography. N-Benzylglycine tert-butyl ester yielded high yields in both reactions with good enantiomeric purity.
