2-Amino-5-iodobenzonitrile is a potent inhibitor of the HER2/EGFR tyrosine kinase. It has shown potent inhibitory activity against mutant and wild type forms of EGFR, including Lapatinib-resistant and -sensitive EGFR mutants. 2-Amino-5-iodobenzonitrile was found to be a selective inhibitor of the EGFR with an IC50 value of 0.1 μM in human lung cancer cells, with no significant effect on normal lung cells. This drug has also been shown to have anti-tumor effects in vivo, inhibiting tumor growth and metastasis in mice bearing xenografts of human lung cancer cells. 2-Amino-5-iodobenzonitrile also inhibits the proliferation of breast cancer cell lines (e.g., MCF7) by binding to the ATP site on the HER2/EGFR heterodimer receptor complex,
