Cyclohexanecarbonyl-L-leucine (CHL) is a new amino acid analog that is orally bioavailable, does not have gastrointestinal side effects, and has a high permeability. CHL has been shown to be readily absorbed into the systemic circulation in rats after oral administration. Mechanistic studies have revealed that CHL is metabolized by the liver, with subsequent uptake by the small intestine. The absorption of CHL was found to be dependent on the presence of α-amino acids in the gut lumen, which may explain its low oral bioavailability.
