Desciclovir is a synthetic antiviral drug that is active against herpes simplex virus. It is also active against cytomegalovirus and varicella-zoster virus. Desciclovir belongs to the class of drugs called nucleoside analogues. It inhibits viral replication by forming a covalent bond with viral DNA polymerase, thereby inhibiting the synthesis of viral DNA. This prevents transcription and replication of the viral genome to produce new viruses. The mechanism of action appears to be unique among currently available antiviral agents, as it does not require activation by cellular enzymes or the presence of intracellular cofactors such as thymidine kinase or ribonucleotide reductase.
