Nelfinavir is an antiretroviral drug that inhibits the protease enzyme. It blocks the proteolytic cleavage of viral proteins, thereby preventing viral replication. Nelfinavir is primarily used to treat HIV-1 infection. It has been shown to be effective in treatment trials involving patients with infectious diseases, such as malaria and tuberculosis. Nelfinavir is a substrate for drug transporters and can interact with other drugs (e.g., ketoconazole). Inhibitors of nelfinavir include α1-acid glycoprotein and genotoxic agents such as anthracyclines and doxorubicin. Mitochondrial membrane potential was shown to be decreased following exposure to nelfinavir, which may contribute to its cytotoxic effect on cells.
