W-7 hydrochloride is a mitochondria-targeted prodrug that inhibits the mitochondrial calcium uniporter (MCU) in cells. This inhibition of the MCU leads to the accumulation of cytosolic calcium, which initiates a cascade of events leading to cell death. W-7 hydrochloride has been shown to inhibit mitochondrial membrane potential and induce neuronal death in vitro. It also induces reactive oxygen species production by mitochondria and causes cardiac dysfunction. Transcriptomic analysis indicates that W-7 hydrochloride affects genes involved in mitochondrial function, such as ATPase, adenosine uptake, and transcription factors. The drug also inhibits DNA synthesis and alters the structure of cell nuclei.
