1,1,2-Triphenyl-2-(4-bromomethylphenyl)ethylene (TPE) is a fluorescent molecule that is used to study protein interactions. This molecule emits light when it absorbs light at an interface and can be used to measure the quantum efficiency of a fluorescence microscope. The emission wavelength of TPE can be changed by altering the length of the alkyl chain. TPE has a quantum efficiency of 0.5% in air and 1% in vacuum and so is more commonly used in fluorescence microscopy than as an analytical tool. Labeling with TPE can be done using cysteine residues on proteins or other molecules. Optimizing TPE for labeling requires changes to the molecule's parameters, such as changing its concentration or pH or adding surfactants to create a hydrophobic environment for labeling.
