Nosiheptide is a thiopeptide antibiotic that is a mixture of (E) and (Z) isomers. It is effective against Gram-positive bacteria, such as staphylococci and streptococci, and Gram-negative bacteria, such as Escherichia coli and Pseudomonas aeruginosa. Nosiheptide binds to the 3-amino-4-chlorobenzoic acid in the bacterial cell wall by competitive inhibition. This binding prevents the formation of an antibiotic-inhibitor complex with the enzyme cell wall synthesis that is required for cell wall biosynthesis, inhibiting protein synthesis and cell division. Lactone ring opens up, forming hydroxymethyl group which acts as hydrogen donor.
