Longipedlactone J is a natural product that belongs to the class of cyclopentenones. It is biosynthesized by a reaction between 3-hydroxy-2,5-dihydropyran and 1,4-diacetylbenzene. The compound has antiviral effects against herpes simplex virus type 1 and 2 (HSV1 and HSV2) and cytomegalovirus (CMV). Longipedlactone J also inhibits the replication of HIV-1 with an IC50 of 0.04 μM in the presence of human serum albumin. The antiviral activity is related to the nucleophilic character of the molecule, which reacts with nucleophiles on viral proteins. This inhibition prevents viral replication by blocking DNA synthesis.
