3’-O-tert-Butyldiphenylsilylthymidine is a synthetic nucleoside that has antiviral activity. It is the first member of a new class of antivirals, and it has been shown to inhibit HIV replication in vitro. 3’-O-tert-Butyldiphenylsilylthymidine binds to the viral RNA polymerase and prevents the synthesis of viral DNA by inhibiting transcription. 3’-O-tert-Butyldiphenylsilylthymidine also inhibits HIV protease, which is an enzyme that cleaves HIV proteins into smaller pieces for the virus to assemble. This drug has not yet been tested in humans.
