4-Amino-2',4'-dichlorodiphenyl ether (4AD) is a diphenyl ether that has been shown to be an activator of the Ah receptor and thus may have potential as a therapeutic agent for the treatment of lung cancer. The mutagenicity of 4AD in a variety of tissues, including human embryonic lung cells, was confirmed in the Ames test. 4AD has also been shown to inhibit the growth of bacteria in emulsions. In long-term studies, 4AD induced teratogenic effects in rats. Pregnant female rats were given doses of 20 and 40 mg/kg/day during gestation days 6 through 15. A statistically significant increase in fetal malformations was observed at both doses. The type and severity of malformations did not appear to be dose related.
