Bradykinin is a peptide hormone that can be formed from the breakdown of kininogen. It works by binding to specific receptors found on the surface of cells and activating a G-protein coupled receptor. Bradykinin stimulates the secretion of gastric acid, increases the permeability of capillaries in the lungs, and causes contraction of smooth muscle in many organs. Bradykinin also has an important role in regulating blood pressure and heart rate. Des-Arg9-[Leu8]-Bradykinin (D-BK) is a potent inhibitor of this activity and is used as an antihypertensive agent. D-BK prevents the activation of G-proteins by binding to receptors with high affinity and specificity. When it binds to its receptor, it inhibits cyclic AMP production, which leads to decreased cell activity and vasodilation. This inhibition reduces blood pressure by reducing cardiac output and increasing systemic vascular resistance.
