Originally isolated from the sub-maxillary glands of mice by Cohen, this synthetic 6kDa human Epidermal Growth Factor (EGF) protein consists of 53 amino acid residues with the intermolecular disulphide bonds: Cys6-Cys20, Cys14-Cys31, and Cys33-Cys42. EGF is a growth factor which serves to stimulate cell growth, proliferation and differentiation by binding to the Epidermal Growth Factor receptor (EGFR).
When EGF binds to EGFR, the receptor undergoes oligomerization with ErbB2/c-neu, ErbB3, and ErbB4 and is phosphorylated. Consequently pathways such as PLC-γ1, PI-3 Kinase, GAP-MAP-raf kinase are activated which play a role in cell proliferation, survival and migration. EGF is also critical in stimulating blastocyst formation and embryo implantation, and further into development it is necessary for the production of the gastrointestinal tract and body mass regulation. Interestingly EGF is also involved in wound healing, and its tyrosine kinase activity increases the growth, metastatization and survival of tumours, particularly in the bone. Studies have shown the EGFR to be expressed in osteosarcoma cells resulting in local metastatization and a worse prognosis.
