FIM-1 is a lipid that is found in the microenvironment of cells. It has been shown to be able to activate the receptor for the tumor necrosis factor, which induces apoptosis and cell death. FIM-1 activates murine leukemia cells as well as human leukemic cells, inducing cellular and necrotic cell death. The gene encoding FIM-1 has been cloned and sequenced. There are two variants of this gene: one in mice and one in humans. The mouse variant contains a factor receptor binding site that can be activated by phorbol esters, such as 12-O-tetradecanoylphorbol 13-acetate (TPA). This site does not exist in the human gene. Thus, TPA cannot activate FIM-1 protein in human cells and cause apoptosis or necrosis.
