Varlitinib is a small molecule inhibitor of the EGF receptor tyrosine kinase with minimal toxicity in animals. Varlitinib inhibits tumor growth and angiogenesis by blocking the binding of EGF to its receptors on epidermal cells, thereby inhibiting the activation of downstream signaling pathways, such as Ras-Raf-MEK-ERK. Varlitinib also has an inhibitory effect on receptor tyrosine kinases other than EGFR, which may be due to its ability to bind to effector proteins that are involved in signal transduction. By blocking these effector proteins, varlitinib prevents cancer cells from responding to signals that stimulate their proliferation and survival.
