Paromomycin is a polypeptide antibiotic that is used to treat infections caused by the protozoan parasite Leishmania. It inhibits amastigotes, which are the intracellular form of leishmania, by binding to cyclohexyl and analogs. In vivo, paromomycin inhibits the growth of leishmanial organisms in mice and rats. This drug also has antileishmanial activity against promastigotes and metacyclic forms of leishmania. Paromomycin binds to phospholipids in the membrane of amastigotes, inhibiting cellular respiration at complex I. The structural similarity between paromomycin and 2-(1,2,3,4-tetrahydronaphthalen-1-yl)-1H-imidazole suggests that they may inhibit the same enzyme or target on the parasite’s cell membrane.
