CDK12-IN-3 is a potent and selective inhibitor of CDK2, CDK4, and CDK6. It has been shown to inhibit the activity of these kinases in vitro, with IC50 values of 0.5 nM for CDK2 and 1.5 nM for CDK4 and CDK6. In vivo studies in mouse models have demonstrated that treatment with CDK12-IN-3 significantly reduced tumor size in lung cancer xenografts. The compound also showed antitumor activity in mice bearing human colon carcinoma xenografts, as well as in an orthotopic model of human prostate cancer. Preliminary results from a Phase I study evaluating the safety, tolerability, and pharmacokinetics of single doses of CDK12-IN-3 have been reported.
