GDC-0575 is a potentiator of colony-stimulating factor (CSF) that has minimal toxicity. It has been shown to enhance the effects of CSF on tumor cells, which may be due to its ability to induce the production of effector proteins. GDC-0575 binds to the catalytic subunit of DNA polymerase δ, blocking DNA replication and inhibiting tumor growth in mice. This agent is also a predictive biomarker for anti-tumor activity because it inhibits myeloid-derived suppressor cells by binding to Chk1 kinase. GDC-0575 has pharmacokinetic properties that are similar to those seen with other CSFs, making it potentially useful for cancer therapy.
