CRA 026440 is a histone deacetylase (HDAC) inhibitor that inhibits the activity of HDACs. It has potent antitumor activity in preclinical models and has been shown to be safe and well tolerated in human clinical trials. CRA 026440 has been shown to significantly inhibit the growth of tumor xenografts in wild-type mice. This agent also induces apoptosis by reducing the expression of pro-inflammatory cytokines such as tumor necrosis factor-α (TNF-α) and interleukin 6 (IL-6). In vitro experiments have shown that CRA 026440 inhibits translation, which may be due to its ability to modulate histone acetylation.
