CP-47,497 is a monophenolic synthetic cannabinoid receptor agonist with a strong affinity for the cannabinoid receptor CB1 (Kd= 2.1 nM)1. Binding to the CB1 receptor triggers inhibition of adenylate cyclase as well as inhibition of N-type voltage-dependent Ca2+ channels and activation of MAPK. In vitro assays showed activation of the caspase pathway and resulting dose-dependent cytotoxicity upon CB1 binding to CP-47,497 in neuroblastoma and glioma cell lines2. In vivo studies on experimental mice, rat and canine models showed depression of locomotory activity, anticonvulsant and hypothermic effects, and strong antinociceptive activity3.
