polymyxine B est un antibiotique polypeptidique cyclique principalement utilisé pour traiter les infections causées par des bactéries Gram-négatives. Il est dérivé de la bactérie Bacillus polymyxa et présente une structure unique caractérisée par une queue hydrophobe et un anneau peptidique chargé positivement, ce qui lui permet de perturber les membranes cellulaires bactériennes. Cette perturbation entraîne une augmentation de la perméabilité et, en fin de compte, une lyse cellulaire, ce qui le rend efficace contre une gamme de pathogènes, y compris Pseudomonas aeruginosa et Escherichia coli. polymyxine B est souvent utilisé dans des formulations topiques en raison de sa néphrotoxicité et de sa neurotoxicité lorsqu'il est administré par voie systémique. On le trouve couramment dans des pommades, des gouttes ophtalmiques et d'autres préparations topiques. Polymyxin B is a sulfonamide antibiotic with broad spectrum of activity against Gram-negative bacteria. It binds to the bacterial membrane, causing an increase in the cell’s permeability and thus leading to cell death. A study on human neutrophils showed that polymyxin B reduces the expression of neomycin sulfate through inhibition of protein synthesis, which leads to cell death by lysosomal enzyme release. This antibiotic has been shown to be effective on clostridium perfringens, where it inhibits bacterial growth and leads to hypersensitivity reactions in some patients.
