2-Keto crizotinib is a potent inhibitor of the tyrosine kinase enzyme, which blocks cancer cell growth. It binds to the ATP site of the enzyme and prevents the phosphorylation of tyrosine residues on cellular proteins, thereby inhibiting protein synthesis. 2-Keto crizotinib has been shown to inhibit receptors that are involved in angiogenesis and metastasis. This agent is also a competitive antagonist at the epidermal growth factor receptor (EGFR) and blocks activation by binding to its extracellular domain. 2-Keto crizotinib also has immunosuppressive effects, which may be due to its ability to block antibody production.
