Trastuzumab

Description :

Trastuzumab, communément connu sous le nom de marque Herceptin, est un anticorps monoclonal utilisé principalement dans le traitement de certains types de cancer du sein et de cancer gastrique qui surexpriment le récepteur 2 du facteur de croissance épidermique humain (HER2). C'est un anticorps IgG1 kappa humanisé dérivé de l'ADN recombinant, ce qui signifie qu'il est conçu pour cibler et se lier spécifiquement à la protéine HER2 à la surface des cellules cancéreuses. Cette liaison inhibe la prolifération de ces cellules et favorise leur destruction par le système immunitaire. Trastuzumab est administré par perfusion intraveineuse et est souvent utilisé en combinaison avec d'autres agents de chimiothérapie pour améliorer l'efficacité thérapeutique. Son mécanisme d'action comprend le blocage des voies de signalisation HER2 et la médiation de la cytotoxicité cellulaire dépendante des anticorps (ADCC). Le médicament est associé à des effets secondaires spécifiques, notamment la cardiotoxicité, ce qui nécessite une surveillance attentive de la fonction cardiaque pendant le traitement. Dans l'ensemble, Trastuzumab représente une avancée significative dans la thérapie ciblée contre le cancer, améliorant les résultats pour les patients atteints de malignités positives pour HER2.

Trastuzumab is a humanised immunoglobulin G1 monoclonal antibody against human epidermal growth factor receptor 2 (HER2), used in breast cancer treatments. Some potential extracellular and intracellular antitumor mechanisms of trastuzumab have been proposed, including activation of antibody-dependent cellular cytotoxicity, inhibition of extracellular domain cleavage, abrogation of intracellular signalling, reduction of angiogenesis and decreased DNA repair that result in tumour cell stasis and/or death. Its drug conjugate, trastuzumab deruxtecan (T-DXd), is used in the clinical therapy for pretreated HER2-mutant non-small cell lung cancer (NSCLC).
