5-Aza-2'-désoxycytidine, également connu sous le nom de décitabine, est un analogue nucléosidique de la cytidine principalement utilisé dans le traitement de certains types de cancer, en particulier les syndromes myélodysplasiques et la leucémie myéloïde aiguë. Sa structure chimique présente une base pyrimidique, spécifiquement une cytosine modifiée, où l'atome d'azote en position 5 est remplacé par un atome de carbone, entraînant l'incorporation d'un groupe méthyle. Cette modification inhibe les méthyltransférases de l'ADN, conduisant à une hypométhylation de l'ADN et à la réactivation de gènes silencieux, ce qui peut induire une différenciation et une apoptose dans les cellules cancéreuses. 5-Aza-2'-désoxycytidine est généralement administré par voie intraveineuse et a une demi-vie relativement courte, nécessitant des protocoles de dosage et d'administration soigneux. Ses effets thérapeutiques sont accompagnés d'effets secondaires potentiels, notamment une myélosuppression et des troubles gastro-intestinaux.
