CSN5i-3 is a small molecule that inhibits the ubiquitin-proteasome system. It has potent antitumor activity in vitro and in vivo, as well as anti-inflammatory effects. CSN5i-3 is a selective inhibitor of the E1 enzyme (ubiquitin ligase) and has been shown to inhibit the proteasomal degradation of prostate cancer cells. CSN5i-3 also has pharmacokinetic properties that are favorable for clinical development and it can be used to treat hepatic steatosis and bone cancer. CSN5i-3 is active against infectious diseases such as malaria, tuberculosis, and HIV by inhibiting host protein synthesis.
